DNA damage occurs as part of normal brain activity, scientists discover

Mar. 24, 2013 ? Scientists at the Gladstone Institutes have discovered that a certain type of DNA damage long thought to be particularly detrimental to brain cells can actually be part of a regular, non-harmful process. The team further found that disruptions to this process occur in mouse models of Alzheimer's disease -- and identified two therapeutic strategies that reduce these disruptions.

Scientists have long known that DNA damage occurs in every cell, accumulating as we age. But a particular type of DNA damage, known as a double-strand break, or DSB, has long been considered a major force behind age-related illnesses such as Alzheimer's. Today, researchers in the laboratory of Gladstone Senior Investigator Lennart Mucke, MD, report in Nature Neuroscience that DSBs in neuronal cells in the brain can also be part of normal brain functions such as learning -- as long as the DSBs are tightly controlled and repaired in good time. Further, the accumulation of the amyloid-beta protein in the brain -- widely thought to be a major cause of Alzheimer's disease -- increases the number of neurons with DSBs and delays their repair.

"It is both novel and intriguing team's finding that the accumulation and repair of DSBs may be part of normal learning," said Fred H. Gage, PhD, of the Salk Institute who was not involved in this study. "Their discovery that the Alzheimer's-like mice exhibited higher baseline DSBs, which weren't repaired, increases these findings' relevance and provides new understanding of this deadly disease's underlying mechanisms."

In laboratory experiments, two groups of mice explored a new environment filled with unfamiliar sights, smells and textures. One group was genetically modified to simulate key aspects of Alzheimer's, and the other was a healthy, control group. As the mice explored, their neurons became stimulated as they processed new information. After two hours, the mice were returned to their familiar, home environment.

The investigators then examined the neurons of the mice for markers of DSBs. The control group showed an increase in DSBs right after they explored the new environment -- but after being returned to their home environment, DSB levels dropped.

"We were initially surprised to find neuronal DSBs in the brains of healthy mice," said Elsa Suberbielle, DVM, PhD, Gladstone postdoctoral fellow and the paper's lead author. "But the close link between neuronal stimulation and DSBs, and the finding that these DSBs were repaired after the mice returned to their home environment, suggest that DSBs are an integral part of normal brain activity. We think that this damage-and-repair pattern might help the animals learn by facilitating rapid changes in the conversion of neuronal DNA into proteins that are involved in forming memories."

The group of mice modified to simulate Alzheimer's had higher DSB levels at the start -- levels that rose even higher during neuronal stimulation. In addition, the team noticed a substantial delay in the DNA-repair process.

To counteract the accumulation of DSBs, the team first used a therapeutic approach built on two recent studies -- one of which was led by Dr. Mucke and his team -- that showed the widely used anti-epileptic drug levetiracetam could improve neuronal communication and memory in both mouse models of Alzheimer's and in humans in the disease's earliest stages. The mice they treated with the FDA-approved drug had fewer DSBs. In their second strategy, they genetically modified mice to lack the brain protein called tau -- another protein implicated in Alzheimer's. This manipulation, which they had previously found to prevent abnormal brain activity, also prevented the excessive accumulation of DSBs.

The team's findings suggest that restoring proper neuronal communication is important for staving off the effects of Alzheimer's -- perhaps by maintaining the delicate balance between DNA damage and repair.

"Currently, we have no effective treatments to slow, prevent or halt Alzheimer's, from which more than 5 million people suffer in the United States alone," said Dr. Mucke, who directs neurological research at Gladstone and is a professor of neuroscience and neurology at the University of California, San Francisco, with which Gladstone is affiliated. "The need to decipher the causes of Alzheimer's and to find better therapeutic solutions has never been more important -- or urgent. Our results suggest that readily available drugs could help protect neurons against some of the damages inflicted by this illness. In the future, we will further explore these therapeutic strategies. We also hope to gain a deeper understanding of the role that DSBs play in learning and memory -- and in the disruption of these important brain functions by Alzheimer's disease."

Other scientists who participated in this research at Gladstone include Pascal Sanchez, PhD, Alexxai Kravitz, PhD, Xin Wang, Kaitlyn Ho, Kirsten Eilertson, PhD, Nino Devidze, PhD, and Anatol Kreitzer, PhD. Two die skydiving in Florida, said to be from Iceland

(Reuters) - Two skydivers from Iceland, one an instructor and the other a student, were killed in a skydiving jump on Saturday, Florida media reported.

The two men were part of a group of 22 skydivers who jumped from a plane in Zephyrhills, about 30 miles northeast of Tampa, according to the Tampa Tribune newspaper.

When only 20 of those who jumped returned, authorities launched a search and found the bodies of the two men a few hours later, Florida media said.

The two men were from Iceland, according to Skydive City, which planned the jump, the Tribune reported.

Their identities were not released.

A spokesman for the Pasco County, Florida Sheriff's Office could not be reached for comment.

(AP) ? Build a state university in the swamplands of south Florida. Move the athletic program to the highest level in college sports and hire a self-made millionaire basketball coach. When his team makes the NCAA tournament, say in Year 2 of its eligibility, beat a tradition-rich opponent like a Georgetown.

That is Florida Gulf Coast University's formula for success.

It took 16 years, and now the school from south Florida is the talk of March Madness.

"Fort Myers is kind of rocking and rolling right now," FGCU forward Eddie Murray said Saturday, less than 24 hours after the No. 15 Eagles beat second-seeded Georgetown 78-68. "They're really excited. This is a big thing for the city and I'm glad we could deliver this."

It was one heck of a delivery.

Suddenly, a school with an enrollment of about 12,000, whose first graduating class wore the caps and gowns in 2001, is getting national attention because of a basketball team loaded with players whose best recruiting offers were from conferences like the Atlantic 10 and Missouri Valley.

The teenagers bought into the pitch from Andy Enfield, a coach made millions starting up a document imaging and contract management company in the health care industry, and who happens to be married to former supermodel Amanda Marcum. They have three children.

Hard to argue with that kind of salesman.

"Yes, we sold this vision," said Enfield, who has also spent time as an assistant coach in the NBA and at Florida State, and has been a shooting consultant for several NBA players. "It wasn't play San Diego State in the (third) round on a Sunday in Philadelphia, it was a vision of success, it was a vision of ... what they could expect in the classroom, off the court and on the basketball court. That's the vision we sell, and I think that's most recruiting. You have to let players know what they should expect from you during their time at school."

Enfield's motto is simple and he's lived up to it.

"I aim for the stars," he said.

Right now he's taking the team and the school with him.

"The biggest thing he's given me has just been confidence," Murray said. "There have been things technique-wise but the biggest thing has definitely been confidence."

The Eagles (25-10) put on a classic performance against Georgetown, putting on an offensive display unlike any seen against the Hoyas this season. Their 21-2 run in the second half stunned and delighted the sellout crowd at the Wells Fargo Center, and their ability to hold off final charge by the Big East team, sold the fans on the legitimacy of the win.

"Early on in the game I kind of thought that more people would be going for the underdog, but they weren't really too vocal in the beginning," Murray said. "As we started getting a couple of dunks here and there, knocking down a couple of 3s and going on a big run against Georgetown, the crowd really started to erupt, become more vocal. The place really started rocking there for a while. Yeah, all these people in Philadelphia have really come to like our team and are rooting for us."

So is a lot of the country, despite the Eagles shattering bracket sheets.

"I've received congratulatory message and advice from other coaches, people in the media, family, friends, people I haven't seen or heard from in 20 years," Enfield said, estimating his phone had 450 text messages ? and counting.

While Fort Myers and nearby Naples may be known for their retirement communities, the area also has some good golf courses, wetland areas ? and the wildlife that comes with them.

"You would see wild animals crossing the road," said Murray, a local who grew up about 20 miles away from the FGCU campus. "Personally, I've seen everything from wild boar, bobcats and, of course, alligators."

That didn't deter the students ? and basketball players ? from attending school on a campus just a few minutes from the Gulf of Mexico that has dorms overlooking a beach on a lake.

"I'm from Orlando, so even being from Florida, a lot of people that I'm close to, they didn't even know where Florida Gulf Coast was or they never heard of it," said Sherwood Brown, the Atlantic Sun player of the year, who led the Eagles against Georgetown with 24 points.

One person who knows something about FGCU is Steve Fisher, the coach of San Diego State (23-10), which will face the Eagles with a berth in the Sweet 16 at stake. A No. 15 seed has never made it to the regional semifinals, while the Aztecs are looking to do it for the second time in three seasons.

"I probably knew more than any coach in America about them because I've got a condo that I've had from my days in the Midwest in Fort Myers Beach, a stone's throw from Florida Gulf Coast," said Fisher, who coached Michigan to the 1989 national championship and two other Final Four appearances.

"I read all about them starting sports. I went over to the campus. I've been on the campus. I've toured it.

"They're good. I think it's legitimate."

Fisher said the low seeding and the short history has nothing to do with this team.

"If we were playing a shirts and skins game with all 64 teams and you brought all the teams out there and watched them warm up, you'd be hard pressed to say, 'Well, this is a team that's not supposed to win,'" he said. "They're good. They're talented. They're well-coached. And they played terrific last night."

The Aztecs broke open their 70-55 win over Oklahoma with a late run behind Jamal Franklin, who leads the San Diego State in scoring, rebounding, assists and steals.

"We can't get ahead to the Elite 8 or the Sweet 16," Franklin said. "Tomorrow is Sunday and that brings Florida Gulf Coast in our path and we've got to worry about playing a good team and we've got to worry about getting that win."

Pope Francis, pope emeritus Benedict XVI meet

CASTEL GANDOLFO, Italy (Reuters) - A reigning pope and a former pope faced each other for the first time in at least 600 years on Saturday when Pope Francis travelled south of Rome for lunch with his predecessor, pope emeritus Benedict XVI.

Francis, who was elected on March 13, flew by helicopter to the papal summer residence at Castel Gandolfo, where Benedict has been living since he abdicated on February 28.

Footage released by the Vatican showed the two men, both dressed in nearly identical white clerical garb, including white skull caps, embracing shortly after the helicopter landed at the large estate.

The only difference in garb is that Francis also wears a short white cape over his cassock and a white sash around his waist - both symbols of his authority. On Saturday Benedict, who often suffered from chills, even indoors, wore a thick white vest over his cassock.

They then rode in the same car to the residence where they prayed together, spoke alone for about 45 minutes and then had lunch accompanied by their two secretaries. Vatican spokesman Father Federico Lombardi said the atmosphere for the entire visit, which lasted just under three hours, was "family-like".

When they went to pray in the chapel, Benedict offered the place of honor, a kneeler before the altar, to Francis, who declined, saying, "We are brothers, we pray together," Lombardi said. Footage showed the two men praying at the same pew.

Asked about the health of Benedict, who became the first pope in 600 years to resign instead of ruling for life, citing diminishing strength, Lombardi said: "It is normal, he is an old man."

Benedict has been living temporarily in the summer residence in the Alban Hills and will move back to the Vatican after the restoration of a convent where he is expected to live for the rest of his life.

Shortly before his resignation, Benedict, now 85, said he would be "withdrawing into prayer" and would live out his remaining days "hidden from the world".

In February, on the last day of his nearly eight years as leader of the Catholic Church, Benedict pledged his unconditional obedience to whoever would succeed him. Lombardi said the meeting "gave Benedict the opportunity, through his gestures" to renew his pledge personally.

While Lombardi said there would be no statement on what the two discussed, it was likely that the conversation included problems of Vatican administration.

LEAKS SCANDAL

Before he resigned, Benedict left a secret report for Francis on the so-called "Vatileaks" scandal in which sensitive papal documents were stolen from the pope's desk and leaked to the media by his butler.

Last year, the butler, Paolo Gabriele, was arrested and sentenced by a Vatican court to 18 months in prison but Benedict pardoned him and he was freed last Christmas.

The presence of a reigning pope and a pope emeritus is new for the Church in the modern era, but experts say it should not cause difficulties unless Benedict tries to influence Francis's decisions, something he has promised not to do.

Some Church scholars worry that in the event that Francis undoes some of Benedict's policies while he is still alive, the former pope could become a lightning rod for conservatives and polarize the Church.

"Benedict XVI could turn into a shadow pope who has stepped down but can still exert indirect influence," Hans Kung, a dissident Swiss theologian who has clashed with Benedict in the past, told a German magazine.

Francis, who inherited a Church riven by problems such as the sexual abuse scandal in many countries, has indicated in his first few days that his papacy will be more austere.

He wants the Church to be poorer and to be closer to the poor and suffering. In this vein, he has decided to hold Holy Thursday service next week in a juvenile jail on Rome's outskirts rather than in the Vatican or in a Rome Basilica, where it has been held by all his predecessor in living memory.
